Manufacturing Process and Market Trends:

The main process for triamcinolone acetonide is a steroidal semi-synthetic route. The mainstream method uses acetate tetraenes or prednisolones as starting materials, followed by dihydroxyl oxidation and bromohydroxyl epoxidation to obtain intermediates. Fluorination is then performed at -40°C using a hydrofluoric acid-acetone system. Subsequently, perchloric acid is added to acetone for isopropylidene ketal protection at the 16-17 positions. After neutralization, decolorization and recrystallization using dichloromethane, methanol, and activated carbon are performed to achieve HPLC purity of over 99%. Crystal form control and residual solvent management are two key points for API compliance with European and American pharmacopoeias. The highly corrosive fluorinating reagents place high demands on equipment materials and waste HF treatment.

The global market is supported by the foundation laid by Sanofi's original drugs. After the patents expired, API production capacity is highly concentrated in China and India. Europe and North America focus on formulation brands and high-end inhalation and injection formulations.
