Visão geral

Scope of the Report

GalNAc is a ligand for the Asialoglycoprotein receptor (ASGPR), which is an endocytic receptor that is highly specifically expressed on the surface of hepatocytes and is almost undetectable in other cell types. ASGPR mediates the clathrin-dependent endocytosis, effectively transporting galactose-derived ligands from the cell surface to the cytoplasm. During this process, ASGPR appears on the cell surface within 15 minutes. Trivalent and tetravalent ligands exhibit higher affinity than monovalent and divalent ASGPR. Therefore, trivalent or tetravalent GalNAc moieties are covalently conjugated to siRNA with proprietary linker structures. In the GalNAc delivery system, GalNAc can bind to small interfering RNA (siRNA) or other therapeutic molecules to form complexes. These complexes enter the cells by binding to ASGPR on the surface of hepatocytes. Given the liver's crucial role in drug metabolism and clearance, this targeted delivery enhances therapeutic efficacy while reducing systemic side effects. The GalNAc delivery system shows great potential in drug development, particularly for treating liver-related diseases, genetic disorders, and certain cancers. By optimizing the chemical structure and conjugation methods of GalNAc, its targeting ability and biocompatibility can be further improved, thereby enhancing therapeutic outcomes.
