It has CAS number 102396-24-7, molecular formula C36H45BrN4O6, and molecular weight of approximately 709.67. Commercial Jasplakinolide is generally supplied as a white, off-white, or pale yellow solid, powder, or film in 50 microgram, 100 microgram, or custom packs. It requires low-temperature, dry, and light-protected storage and is commonly prepared in DMSO, ethanol, or methanol before use. Structurally, it contains a brominated indole unit, phenolic functionality, multiple amide bonds, and a lactone-containing macrocyclic scaffold. It is classified as a cell-permeable actin modulator. Manufacturing requires stringent stereochemical control, efficient macrocyclization, impurity removal, isomer control, and analytical confirmation by HPLC, MS, and NMR. Its mechanism is to bind filamentous actin at a site overlapping with phalloidin, promote actin nucleation and polymerization, and stabilize pre-existing actin filaments. It is mainly used in cytoskeleton biology, cell migration, endocytosis and exocytosis, neurodevelopment, cancer invasion, stem-cell differentiation, and live-cell imaging probe development, and is intended for research use only.
