Nitrilase is a type of biocatalytic enzyme that directly hydrolyzes the —CN group in nitrile compounds to produce carboxylic acids and ammonia, and is an important category within nitrile-converting enzymes. Compared with conventional strong acid, strong alkali, or high-temperature hydrolysis processes, nitrilase enables milder reaction conditions and can usually operate in aqueous systems under neutral, weakly acidic, or weakly alkaline conditions. It offers advantages such as high selectivity, fewer side reactions, and lower environmental burden. Due to differences in enzyme source and engineering modification, different nitrilases vary significantly in substrate scope, chiral selectivity, regioselectivity, thermal stability, and pH tolerance. Commercial products are generally supplied in forms such as free enzyme preparation, immobilized enzyme preparation, and whole-cell catalyst.

The upstream of nitrilase mainly includes microbial strain resources, gene sequences, expression vectors, fermentation media, enzyme engineering technologies, immobilization carriers, and enzyme formulation excipients. The midstream involves enzyme screening, directed evolution, fermentation expression, purification, immobilization treatment, and process scale-up. Downstream applications are mainly concentrated in pharmaceutical biocatalysis, agrochemical intermediate, specialty fine chemical, and environmental remediation. In pharmaceutical and CDMO applications, nitrilase can be used for the green synthesis of chiral carboxylic acids, aromatic carboxylic acids, heterocyclic carboxylic acids, and other API intermediates. In agrochemical and specialty fine chemical applications, it is used for selective transformation of nitrile-containing substrates, replacing part of traditional chemical hydrolysis routes. In environmental remediation, nitrilase can be used for nitrile wastewater treatment and degradation of nitrile-containing pollutants.
